I'd go with Blaster Master personally... if we're talking strictly about the NES (noticing a few people referencing SNES games) - though there definitely were a lot of great gems (for me the franchise titles like Metroid, Castlevania, Zelda, etc. were just 'consistently solid', not necessarily 'the best games on the system' - as most came into their own right on later systems with Super Metroid, SoTN, Link to the Past, etc. and have been boosted by the nostalgia factor).

Though a couple honorable mentions no one seems to mention: Guerilla War and Blades of Steel

Kirby's Adventure has to be my favourite, as it's the game that got me hooked on the Kirby series in the first place.
I'm winding up my jack-in-the-box, but the only thing popping out is nuclear warfare.

I'm actually gonna go out on a limb here with one I know will be an unpopular vote:

    Final Fantasy II

Before you correct me, I do not mean FFIV's US release for SNES, I mean FF2 for Famicom. It proved that the NES could handle an RPG with a developed story and characters, introduced the ability to do a "code word" system, provided the concept for non-fixed levels to RPGs, and moved away from Vancian/D&D magic to a Mana-based system. Sadly, it was "too much, too soon", but FF2 remains a really ambitious game which deserves more respect and attention. While 1 and 3 are more playable games, I feel that FF2 punches well above its weight and is ignored in favor of safer games.

Outside of that, there's a real obvious one for me, Mega Man 6. The final of the Mega Man Classic Series, this game is (hopefully) fondly remembered by all fans as the capstone to a great series. (7+ I consider the "Neo-Classic" series) The Classic Series developed the basic concepts of the Super Fighting Robot, Mega Man, his Mega Buster, his family, and his world. Everything after built on that strong foundation, and I'm glad that it developed so well on the NES, the Game Boy, and then onwards.
