I have played all 32 time trial courses but Toadette still hasn't been unlocked? I think I have beaten 30/32 courses, do I need to beat all of them?

Please, next time provide more info on what game are you playing. Since you didn't mentioned it in your post, I assumed it's Mario Kart Wii. Have a look at this.
